Is there any problem using 6s and a 4s to make 10s? I'm in the process of building a mhz mystic with twin 1527 neu's

No problem as long as all the cells are the same make, C rating and capacity. Actually it is pretty common for guys wanting to use existing 4S and 6S packs instead of buying 5S packs.

Yes, no problem combing packs in series to make a larger single pack so long as you follow what Jay mentioned.
